Talk to me about… your research

I am a Professor in Biostatistics at the Harvard T.H. Chan School of Public Health and an Associate Member at the Broad Institute.  I participated extensively in the NIH Human Microbiome Project and co-leads the "HMP2" Center for Characterizing the gut microbial ecosystem in IBD and the Human Microbiome Bioactives Resource.  My  lab focuses on computational methods for functional analysis of microbial communities.  This includes systems biology reconstructions integrating metagenomic, metatranscriptomic, and other microbial community 'omics, the human microbiome in autoimmune disease such as IBD, and its potential as a biomarker and route for therapeutic intervention.

 

Talk to me about…something people may not know about you

I'm a happy kitty parent, most recently of two new kittens who (like human babies) have as yet to learn to sleep through the night. I wish I had as much energy as they do!

He is the co-founder of several additional startup companies, including a Connecticut-based microbiome company, Azitra Inc. He brings strong background in entrepreneurship and business, and is also a partner in a venture capital fund, Bios Partners, bringing experience in public markets, drug development, and venture capital investments.

Our mission at Siolta Therapeutics is to transform the therapeutic landscape of inflammatory diseases, such as asthma, through the power of the microbiome. As the lead executive driving Siolta’s early-stage development, I head a talented team of scientists, blending microbiology, immunology, and bioinformatics expertise to leverage microbiome data for the improvement of patient stratification and development of targeted microbial therapeutics. We are developing FDA-approved live biotherapeutics, as well as companion diagnostics, aimed not only at treating disease but also preventing disease, starting with asthma.

My two professional passions have always been environmental systems and health. I have spent the entirety of my career exploring the relationship between these two concepts, from my time in the Solomon Islands as a Peace Corps Volunteer to my PhD work in marine ecosystems impacted by environmental alterations.

Dynamic microbial and viral communities constantly inhabit our bodies, encoding the majority of the unique genes that alter these processes. Resident microbial and viral community composition is unique to each human.

Specifically, I am working to understand the role of persistent microbial colonization in triggering inflammatory episodes in Cystic Fibrosis patients. Understanding microbial interactions and changes in microbial physiology that are associated with changes in patient status may lead to the development of biomarkers to diagnose changes in infection earlier and more specifically.

I am also interested in how phage shape dynamic host-associated microbial communities, and in characterizing the function of the 70% of phage encoded genes with no known function.

I am the the leader of the Complex Microbial Systems Group at the National Institute of Standards and Technology (NIST) in Gaithersburg, MD. My current role involves leading international efforts to improve microbiome and metagenomic measurements by organizing inter-lab studies, developing reference materials and reference methods, and developing in vitro tools that allow us to better understand microbial community resilience and evolution Prior to joining NIST in 2014, I spent 11 years as a principal investigator with the FDA where I focused on characterizing the global genomic diversity of enteric pathogens, with applications for food safety, bioforensics and public health.

I am the chief scientific officer at Da Volterra, a French biotechnology company, headquartered in Paris, France. Da Volterra is committed to the discovery and development of innovative approaches to prevent the deleterious consequences of antibiotic treatments on the intestinal microbiota, including the emer­gence and dissemination of antimicrobial resistance, as well as the occurrence of infections by opportunistic pathogens such as Clostridium difficile. Prior to that, I led an academic career as a cell biologist focusing on signal transduction and oncogenesis, ultimately leading a research laboratory at the Institut Curie in Paris.

 

Talk to me about…something people may not know about you

I was born in Canada, lived for most of my life in France, and am now based in the London. I am an avid off-piste skier, in the French and Swiss Alps. I particularly enjoy the slow hiking/skinning up the mountains which gives me time to appreciate the fabulous scenery, and to think in depth about new and arising matters, in particular in relation to my research.

Prior to joining Seres, he served as the Chief Operating Officer for the International AIDS Vaccine Initiative (IAVI), a global R&D organization. Prior to IAVI, Dr. Cook was the founding CEO at Anza Therapeutics, a biotechnology startup that focused on developing a microbial vaccine platform to induce cellular immune responses. Before launching Anza, Dr. Cook held positions of increasing responsibility at the biotechnology corporations Cerus and Eligix, overseeing R&D, program management, manufacturing, and clinical and regulatory affairs. He has led teams in the development and commercialization of several biotech products and has been directly responsible for obtaining marketing authorization from the European Union for four medical products. Dr. Cook is also a co-inventor on more than 25 patents. He earned his undergraduate degree from Harvard College and his Ph.D. in chemistry from the University of California, Berkeley.

I studied chemistry at the VU University Amsterdam, where I progressed to obtain a PhD in biophysics from the same university.

Tell me something about…your research

I am a senior postdoctoral fellow at Don Ingber’s lab at the Wyss Institute for Biologically Inspired Engineering at Harvard University.I received my PhD from the Hebrew University of Jerusalem and is a Hoffman Leadership and Responsibility Fellowship Program laureate. Tell me something about…your research

I am a senior postdoctoral fellow at Don Ingber’s lab at the Wyss Institute for Biologically Inspired Engineering at Harvard University.I received my PhD from the Hebrew University of Jerusalem and is a Hoffman Leadership and Responsibility Fellowship Program laureate. I am a certified dietitian with past in/out patient and private clinic experience in clinical nutrition.

I have extensive research experience in intestinal physiology and pathophysiology with a focus on mechanisms of gut inflammation. My biophysical work covers the development of a label free SPR based cells biosensor and a method for cryopreservation of adherent cells.

My current work at the Ingber lab focuses on developing in-vitro models for intestinal infection and Environmental Enteric Dysfunction (EED) using Organ-On-Chip microfluidic technology that can recapitulate the multicellular architectures, tissue-tissue interfaces, chemical gradients, mechanical cues and vascular perfusion of the body.

Brad is a Project Director in Immunology Discovery.  He has led both internal drug discovery projects and external collaborations including work on the microbiome with a focus on therapeutics for the treatment of IBD.  He leads a cross-functional group within Abbvie responsible for executing on the Discovery and Clinical strategy in Gastroenterology.  His experience in project leadership, immunology, and in vivo pharmacology ranges from the target identification phase through drug candidate selection and IND.
